Overview
iPSCs Facility is located on the 5th floor, South Wing of the Vallisneri Building, rooms 14, 15 and 27.

The facility consists of five separate rooms. Room 1 is equipped with one laminar flow hood, one CO2/N2 incubator, one centrifuge, one refrigerator (+4 and -20 C°), and one water bath. Room 2 is equipped with one laminar flow hood with two working stations. Room 3 is equipped with one CO2 incubator. Room 4 is equipped with a Nikon inverted fluorescence microscope, a laminar flow hood and room 5 is equipped with a liquid nitrogen tank and refrigerator (+4 and -20 C°).
The hoods and the incubators are exclusively dedicated to work with cultures of pluripotent stem cells (maintenance and differentiation). The microscope is equipped with a camera, and three fluorescence filters (560/470/285 LED units). It can be used by all users for routine monitoring of iPSC cultures, furthermore it will be possible to book longer slots for fluorescence acquisition. Liquid nitrogen tank is provided for storage of frozen cells. Two refrigerators are for the storage of medium, supplements, citokines and other reagents needed for the work with iPSCs.
Guidelines
Access to the facility is possible only with the authorization of the facility manager and after training from the facility technician.
Prior to their use in the facility, all PSC/iPSC lines must be tested for mycoplasma. The facility can be used for any type of PSC cells differentiation and maintenance of their derivatives.
New users must be specifically trained before working in the facility.
The specific training includes the study of the documentation provided by the Facility on the
handling of the equipment, safety rules and general rules of maintaining the culture of PSCs.
These training sessions are held by appointment. The candidate user should contact the Facility technician by e-mail (evgenii.lysenko@unipd.it) for documentation and the appointment.
Users cannot work in the iPSCs Facility unless all the training requirements have been met.
Access to the Facility is restricted to those people whose presence is required for experimental or support purposes. The Facility managers and technicians have the final responsibility of assessing each circumstance and deciding who may enter or work in the laboratory.
The room is accessible from Monday to Friday 8:00 a.m. to 6:00 p.m. If the researcher needs to access the laboratory at different hours or on holidays, Facility manager must be informed.
The access to the Facility is subject to reservation using the booking system at
https://service.biologia.unipd.it/gestione_dipartimento/login with username and password. This is essential for the safety of all users. Moreover, the logbook will be used for the calculation of the expenses.
It is mandatory that all personnel wear PPE (personal protective equipment) i.e. lab coats and gloves. Lab coats will be sent for cleaning every 3 weeks. All personnel must wash their hands after removing gloves. Eating, drinking, and storing food is not allowed.
Clean and sanitize with 70% ethanol anything that has been touched before leaving the room.
