GUIDELINES FOR USE OF THE BIOLOGY DEPARTMENT’S CENTRIFUGES

HOW TO USE:
1. Only operates that have been trained can use departmental centrifuges. This training must be done and certificated by Service Manager (Dott. Pavanello). Every user must advise the Service Manager if hasn’t done this training, so that Manager con organize it
2. Every user must reserve by connecting on site service.biologia.unipd.it/gestione_dipartimento/login, and go to the folder PRENOTAZIONE STRUMENTI►Strumento►Servizio Centrifughe, where there are 3 centrifuges loceted at VSUD:
a. Allegra 25R 3 rotors:
- TS5.1 for 15 ml and 50 ml falcon tubes,
- TA14-50 for 50 ml NO falcon tubes
- TA10-250 for 250 ml tubes
b. Avanti J25 3 rotors:
- JA 25.50 for 50 ml NO falcon tubes
- JLA 10-500 for 500 ml tubes
c. Avanti J26 uses the same AvntiJ25 rotors, but it’s out of use, because it is not under maintenance contract
To reserve Ultra LE80K it has go to the folder PRENOTAZIONE STRUMENTI►Strumento► BLS2 Virus Facility, so to book, it must be authorized to use the room BLS2: The service has 3 ultra rotors:
a.70Ti ONLY FOR VIRUS, fixed angle rotor for 30ml tubes, it can reach the maximum speed of 35000rpm. It is found in a refrigerator at 4°C, located inside the BLS2 room and cannot be taken out of it. Is the only one that can be used with VIRUSES
b.70 Ti, fixed angle rotor, for 30 ml tubes, located in a 4°C refrigeretion room at VI NORD floor, NOT FOR USE WITH VIRUSES. There are adapters (P/N 303307) that can be used with the following part numbers tubes:
•Ultra-clear 344085 da 13,5 ml
•thinwall polypropylene 326814 da 13.5 ml
•Thickwall polypropylene 355640 da 10 ml
•Thickwall polycarbonate 355630 da 10 mL
3. Near every centrifuge there is the USE REGISTER, where it must be specifying:
•date
•rotor
•velocity (RPM OR XG)
•temperature
•minutes of use
•user’s name and lab’s telephone number (USE BLOCK LETTERS)
•supervisor’s name of the research group (WRITE FULL NAME WITH BLOCK LETTERS)
4. Falcon tubes (15 ml and 50ml) can be centrifuged only with Allegra 25R’s TS5.1-500 rotor with own adapters
5. Tubes must be closed safely. Tubes should not be filled to the brim, the volume is given in the instructions provided by the manufacturer
6. When the rotors JLA10.500, TS5.1 are used IT MUST KEPT ALL BUCKETS, EVEN IF SOME AREN’T USE
7. Before goes away from centrifuges, the user must be sure that the rotor has arrived at right velocity
8. After use, switch off the centrifuge and leave open the lid to allow to the chamber dry. The rotors after washed and dried, must be put in order:
•Avant Js and Allegra’s rotors in fridge of room 25 at the VSUD
•Ultra’s rotors in the cooling room at the VI NORD
9. The lid’s pin must be spread with spinkote, a lubricant for metal thread that is in the
white cabinet in the room 25 of the V SUD.
10. Check that all rotor’s o-rings are in the right position; JLA10-500 must be placed
inside the closure of each cannister. The thread of cannister must be spread with
spinkote.
SAFETY:
1.It must be notify to the Service’s Manager in case of any malfunction or problems it
has been found on centrifuges before, during or after use
2.It is forbidden to make any intervention on machine or its electric system
3.Do not wear gloves to pick up the rotors and set the centrifuges
4.In the department’s centrifuges is prohibited to centrifuge pathogenic organism and
biological material which may be hazardous
5.The centrifuge tube must be made of glass thick or it’s better if plastic tubes, and they
must be checked before use
6.The buckets must be match for weight, and with tubes correctly balanced
7.It must be used deionized water or alcohol (propanol 70%) to balance empty tubes. IT’S FORBIDDEN USE SODIUM HYPOCLORITE OR PHYSIOLOGICAL SOUTION TO BALANCE, BECOUSE THEY ARE CORROSIVE
8.Always clean centrifuge’s internal chamber, while the rotors must be washed and dryed; after that they must be spread with spinkote, a lubricant for metal thread that is in the white cabinet in the room 25 of the V SUD.
The spinkote must be spread on:
•the pin of the lids of rotor
•the cannister’s threads of rotor JLA 10-500
•on the buckets’s threads of SW28Ti rotors
9.The dirty paper and gloves must be trashed in own laboratory, in the appropriate biological or chemical waste bin
TROUBLESHOOTING:
1.It must notify to the Service’s Manager in case of fault or any problems it has been found to centrifuges
2.If there are vibrations or anomalous rumors, immediately stop the run and call Service’s Manager
3.In case of damaged tubes and loss of material during the run, stop the run and wait at least 30 minutes to allow the deposition of aerosol’s particulate. If the breaking has been detected when the run has finished, it must be change rotor, and it must be leave closed at least 30 minutes.
4.It must wear appropriate DPI for all operations (for example resistant rubber gloves and use forceps to remove glasses or plastic’s fragment)
5.All damaged tubes, glasses fragment, accessories and rotor must be submerged into a disinfectant solution for 24 hours; use to disinfect ethanol 70%
6.It must clean the centrifuge’s internal chamber and leave to act the disinfectant solution overnight, and therefore wash and dry
7.All biological waste must be treated as contaminated material
